Evaluation of Expression of EGFR, HER-2 and COX-2 in Colorectal Cancer

چکیده مقاله:
Background and objective: EGFR and HER-2 are two members of ERbB/HER family of Type I Transmembrane growth factor receptors. Cox2 is an enzyme responsible for the conversion of arachidonic acid to prostaglandins, which has a major role in angiogenesis and can modelate tumor growth. The aim of this study was to determine the level of expression of EGFR, HER-2 and Cox2 in colorectd cancer. Material and Methods: IHC study was performed in paraffin-embedded blocks of 47 patients underwent colectomy due to colorectal cancer in Modarres Hospital, Tehran, Iran from 2008 to 2009. Three separated pathologists analyzed the slides after complete IHC staining for EGFR, HER-2 and COX-2. Results: EGFR, HER-2 and Cox2 revealed over expression in colorectal cancer as 80.9%, 25.5% and 72.4% respectively, EGFR revealed no statistically significant association with clinicopathologic parameters, but Cox2 overexpression exhibited statistically significant association with higher stages tumors (III, IV) (P value: 0.037) and tumor with lymph node metastasis(P= 0.005). On the other hand, HER2 overexpression showed statistically significant association with lower grade (well and moderately differentiation) tumors (P= 0.042). Conclusion: According to over expression of three markers, EGFR, HER-2, and COX-2 in colorectal cancers, using drugs that act against these receptors and investigation of survival improvement of patients with these drugs in other studies are recommended.

متن کاملClinical role of HER-2/neu expression in colorectal cancer.
PURPOSE To evaluate the HER-2/neu expression and its relationship with clinicopathological parameters and prognosis in colorectal cancer patients. METHODS A total of 51 colorectal cancer patients who underwent resection with curative intent from January 2005 to March 2006 were included in this study. Patients were regularly followed up and survival data were obtained as of as April 2011. HER-...
متن کاملOver-expression of COX-2 mRNA in colorectal cancer
BACKGROUND Cyclooxygenase-2 (COX-2, PTGS2) is an enzyme involved in the synthesis of prostaglandins and thromboxanes, which are regulators of biologic processes such as inflammation, cell proliferation and angiogenesis. COX-2 over-expression was reported in many (pre) malignant tissues, but data strongly vary and seem to depend on the methodology used. METHODS Normal colorectal mucosa and pai...
متن کاملHistopathological Evaluation and Analysis of Immunohistochemical Expression of Bcl-2 Oncoprotein in Colorectal Carcinoma
Abstract Introduction: Colorectal cancer is the third most prevalent malignancy with high mortality rate, necessitating markers that predict survival and guide the treatment. Previous studies have examined the immunohistochemical expression of Bcl-2, an apoptotic marker, in colorectal carcinoma, but results have been contradictory. Objectives: To evaluate the histopathological featu...
متن کاملRole of Cyclooxygenese-2 (COX-2) Expression in Breast Cancer Differentiation and Its Relationship with Hormone Receptors Status
Background and Objectives: Cyclooxygenese-2 (COX-2) expression in breast cancer and its correlation with tumor prognosis is unclear. We investigated the incidence of COX-2 expression in patients and assessed interactions between COX-2 and clinical features of cancer and expression of HER2/neu, estrogen receptor (ER), and progesterone receptor (PR). Methods: COX-2 expression was investigated by...
متن کاملHER-2 and HER-3 expression in liver metastases of patients with colorectal cancer
OBJECTIVE In this study, we evaluate the frequency of HER-2 and HER-3 expression in liver metastases from patients with colorectal cancer (CRLM). We analyzed the potential of HER-2 and HER-3 as therapeutic targets and evaluated their prognostic value. PATIENTS AND METHODS Overall 208 patients with CRLM were enrolled. HER-2 and HER-3 expression were determined in metastatic tissue of diagnosti...
